- W955078012 abstract "An important step in the analysis of the molecular structure, function, and morphogenesis of rod cell outer segment (ROS) is the development of preparative procedures to purify intact ROS from retinal tissue extracts, separate the soluble or cytosolic fraction from the ROS membrane, and isolate the disk and plasma membranes. This chapter describes the methods necessary to purify and fractionate ROS and explains the compositional analysis of defined ROS fractions. A number of methods have been developed to isolate ROS from rod-dominant retinas and, in particular, from bovine and frog retinas. Although sucrose gradients have been most widely used, other gradients, such as Ficoll-sucrose, have also produced relatively pure and intact ROS. For frog ROS preparations, Percoli gradient centrifugation is particularly effective to isolate the intact ROS. The chapter also describes the separation of ROS into soluble and membrane fractions." @default.
