- W959177410 abstract "Publisher SummaryThe proliferation of smooth muscle cells is fundamental in the onset of accelerated atherosclerosis, after cardiac transplantation and restenosis. Multiple pathways are involved in the proliferative response of smooth muscle cell to an extracellular stimulus, which has rendered is difficult to identify precise targets of antiproliferative agents. One of the important elements in signal-transduction cascades is protein kinase C, and its inhibition appears to be sufficient to cause the inhibition of smooth muscle cell proliferation in vitro. In this study, a specific inhibition of proliferation in vitro of smooth muscle cells by α-tocopherol at physiological concentrations was observed. Of particular relevance was the finding of an antiproliferative effect of α-tocopherol on rat and human smooth muscle cells. The inhibition of smooth muscle cell proliferation in vitro by α- tocopherol at physiological concentrations may explain the finding that in vivo smooth muscle cell only proliferates under stress situations. β-Tocopherol shows only a minor effect at the level of cell proliferation or protein kinase C activity. Because β-tocopherol prevents the effects of α-tocopherol, it seems justifiable to conclude that the mechanism of action of α-tocopherol as a regulator of smooth muscle cell proliferation cannot be associated with its antioxidant properties. The effects α-tocopherol are indicative of a site-directed recognition mechanism involving a series of events, including the binding of α-tocopherol to a “receptor protein,” activation (or expression) of a protein phosphatase, dephosphorylation of protein kinase, and inhibition of protein kinase C activity, along with affecting gene-transcription inhibition of cell proliferation. These studies provide a molecular interpretation of the epidemiological information linking a decrease of plasma α-tocopherol with an increased risk of ischemic heart disease." @default.
