- W960686591 abstract "This chapter discusses the assay, purification, and properties of δ-aminolevulinic acid (ALA) synthase. ALA synthase is the first enzyme of the porphyrin biosynthetic pathway. Its level in any cell is apparently strictly controlled, and in most tissues, it is low or undetectable. The only cells reported to have high levels of the enzyme are reticulocytes and photosynthetic bacterium, Rhodopseudomonas spheroides. In colorimetric assay methods for ALA synthase, ALA is condensed with acetyl acetone to give a pyrrole, which is measured by the use of Ehrlich reagent. The procedure must include the separation of the related aminoketone aminoacetone. The enzyme is assayed by measuring the conversion of succinate-14C to ALA. Bacterial succinic thiokinase is added to ensure the conversion of succinate to succinyl-CoA. In crude preparations, malonate and antimycin A are added to minimize succinate oxidation, and malate is added to trap any malate-14C formed. The mitochondrial enzyme is unstable, and rapid freezing is essential before it is stored at -15°. Once frozen and thawed, preparations show greater activity than freshly prepared mitochondria, but subsequent freezing and thawing results in a 30–50% loss of activity." @default.
