- W9628118 abstract "Prostatic cancer is the second most common malignancy in American males and the majority of patients are found to have advanced stages of the disease at the time of its discovery [1, 2]. Since the discovery by Huggins and Hodges that prostatic cancer is an androgen-dependent tumor the treatment of advanced prostatic cancer has centered on methods designed to decrease circulating levels of androgen [3]. The majority of patients so treated, either by castration or by the administration of pharmacologic doses of estrogens, display both subjective and objective evidence of tumor regression, but the duration of response to these modes of therapy is variable [4]. Ten percent of patients with metastatic prostatic cancer treated by hormonal manipulations survive less than one year, 50% survive less than three years, and 10% survive more than ten years [5]. Alternative methods of treatment such as chemotherapy are usually reserved for patients who fail to respond to a trial of hormonal therapy [6]. Thus, if one could identify those patients unlikely to obtain a prolonged response to hormonal manipulation they could be spared the side-effects of an unsuccessful course of hormonal therapy and instead be started earlier on alternate modes of therapy at a time when they are most likely to tolerate and thus possibly more likely to respond to these treatment. While a variety of clinical, histological, and biochemical parameters have been studied to predict the biological response of prostatic cancer none have proven completely satisfactory [7–9]." @default.
