- W964945017 abstract "This chapter reviews the principles of large zone size exclusion chromatography (large zone SEC) as applied to the investigation of self-association of macromolecules, specifically proteins. This self-association is not only of fundamental scientific interest, but also has practical consequences with respect to protein purification and to maintenance of biological function. The recent development of recombinant DNA technology has made available substantial amounts of some proteins that were formerly available in very limited amounts. These recombinant proteins must be obtained in a state that is fully biologically active. In some cases proteins are active as monomers, but in others the active protein exists as a dimer or an oligomer. A variety of approaches are used to study the phenomenon of protein self-association. One approach is to dissociate an oligomeric protein by such means as detergents, pH adjustment, addition of salts, or by chemical modifications. Once dissociated, the resulting monomeric subunits can be characterized by determining the molecular weight. Another approach is to study the equilibrium that exists among complexes of various sizes in solution. Such physical methods as sedimentation equilibrium, analytical ultracentrifugation, calorimetry, and concentration difference spectrophotometry are used." @default.
