Certain classes and subclasses of antibodies, when bound to their respective specific antigens, will bind the Clq subcomponent of the C 1 macromolecular complex of complement. Methods for the detection of cell-bound antibodies generally rely either on the binding of anti- immunoglobulins or on the ability of the antigen-antibody complexes to bind complement and initiate the classical pathway that leads to cell lysis. The unbound material, containing Clq, is concentrated to 20 ml by dialysis against polyethylene glycol 6000 then dialyzed against 0.093 M sodium phosphate buffer, pH 5.3, containing 0.2 M NaC1 and applied to a 4.4 cm × 90 cm column of sepharose G-200 equilibrated with the same buffer. Doubling dilutions of test or control sera are prepared in tissue culture medium containing 10%, fetal calf serum (FCS), that has been adjusted at 22 ° to a specific conductivity of 8.3 mmho/cm2 by dilution with 9.25% sucrose. It is found that the two-step procedure can be used to monitor the antibody turnover at the cell surface, where prolonged incubation times may be involved." @default.
