The metabolism of itaconate and mesaconate in animal liver mitochondria is also discussed. A soluble protein fraction from mitochondria is capable of degrading itaconate when suitably fortified. Itaconate and mesaconate are converted by succinic thiokinase to their corresponding coenzyme A thioesters. The enzyme methyl glutaconase converts these two compounds to citramalyl coenzyme A to yield an equilibrium mixture of 13% itaconyl-CoA, 58% citramalyl-CoA, and 29% mesaconyl-CoA at 20 ° and pH 7.4. Citramalyl-CoA is cleaved by an enzyme in the mitochondrial extract to form acetyl-CoA and pyruvate. Several microorganisms catalyze transformations of itaconate, mesaconate, citramalate, or methyl succinate. Pseudomonas species are found to contain a transferase that converts itaconate and citramalate to their respective coenzyme. Itaconate-grown Pseudomonas species also oxidize methyl succinate (both D and L isomers) to mesaeonate, which is converted to pyruvate and acetyl-CoA via citramalate and citramalyl-CoA." @default.
