- W967570522 abstract "Munkank soran kimutattuk, kismolsulyu hősokk-feherjek (sHSP-k) kardioprotektiv hatassal rendelkeznek transzgenikus eger modellen. A hsp27 feherjet overexpresszalo egervonalakban a tumorellenes szerrel (doxorubicin) tortenő kezeles karos mellekhatasa jelentősen csokkent. A kvantitativ RT-PCR kiserletek soran nyert adatok arra utalnak, hogy az sHSP-t overexpresszalo sejtekben a karosodott feherjek lebontasat vegző genek kifejeződese csokken, ami erősen alatamasztja az sHSP-k citoprotektiv szerepet. Kiserleteink ravilagitottak arra is, hogy a stressz-indukalt HSP expresszioban kulcsszerepet jatszhat a membrankotott Rac1 altal medialt jelatviteli mechanizmus is. Ezen eredmenyek elősegithetik uj, membranaktiv gyogyszerek fejleszteset is. Az sHSP-k vedőszerepet szerepet demonstraltuk hasado elesztő (S. pombe) eseteben is. Mice overexpressing hsp27 and treated with the anti-tumor agent (doxorubicin) showed significantly reduced adverse side effects compared to wild type mice. Quantitative RT-PCR data showed that in the sHSP-overexpressing cells the expression of genes involved in protein degradation has been significantly reduced, strongly supporting the cytoprotective role of sHSPs .Our experiments revealed that membrane bound Rac1 might play a key role in mediating the expression of HSPs These results may facilitate the development of new membrane–active drugs. We have also demonstrated the protective role of sHSPs using the fission yeast (S. pombe) as a model. We have shown that elimination of the two sHSPs (despite the presence of trehalose) in the micromammal affect cell membrane composition, anticipating the applicability of the model in the field of drug discovery as well." @default.
