- W970012429 abstract "Publisher Summary This chapter discusses the procedures used in radiation inactivation experiments and the reasons for technical steps. With each refinement, accuracy, reproducibility and validity can increase. Moreover, comparison with known molecular sizes indicates that with the present techniques often attain agreement within 10%; a survey of the older literature indicated agreement better than 15%. The technique of radiation inactivation depends on the deposition of large amounts of energy directly in the biologically active structure under conditions where radiation products from other molecules are without effect on the measured property. Such direct radiation damage is independent of the surrounding environment. It follows that purification is not required; if the starting material has sufficient activity, molecular weights can be determined after irradiation of intact cells, seeds or frozen tissues. Further, certain caveats should always be borne in mind. This method reveals the mass of a functional unit as it exists in the physical state determined by sample preparation and irradiation conditions. The meaning of a functional unit depends on the particular assay which is used. And finally, the target size is a sum of all the necessary parts." @default.
