- W974641890 abstract "This chapter discusses the isolation of physiologically responsive secretory granules from exocrine tissues. Useful criterion is the behavior of isolated secretory granules suspended in physiological saline solutions at the appropriate pH, temperature, and ionic strength. In vivozymogen granules concentrate, store, and subsequently release the macromolecular product into the lumen, as many histological studies have demonstrated. Therefore, it is reasonable that in the absence of exocytotic stimuli, isolated secretory granules should not lyse or release the secretory product when incubated in artificial solutions with compositions similar to the cytosol. With that in mind, this granule isolation technique is designed to provide granules that readily tolerate physiological salt solutions yet remain osmotically active and physiologically responsive to such intracellular messengers as calcium, pH, and phosphorylation." @default.
