First, the amplitude of the hormone pulses in hepatic portal blood is two- to three-fold higher than that in the peripheral circulation. The data from the tracer analyses of the signal can also serve for more definitive kinetic analysis of unit responses to a single hormone pulse using the technique of deconvolution. Deconvolution is a mathematical method for separating the contribution of the physical system from the total response to better estimate the parameters of the biological response. To maintain sterility, the perifusion columns and tubing are gas-sterilized and sterile tissue culture media are used. The new system consists of three siliconized, gas-sterilized, low-pressure chromatography columns perifused in parallel with oxygenated medium. The attached cells and beads from each column are immediately expressed with a syringe of phosphate-buffered serum from the column into a sterile 50-ml tube." @default.
