- W97673770 abstract "This chapter describes the clonal analysis of human natural killer (NK) cells. Cells cultured in interleukin-2 (IL-2) show heterogeneous effector functions in cytotoxicity assays. Specific T-cell lysis in appropriately sensitized cultures, natural killing, and antibody-dependent cellular cytotoxicity are all readily detectable. It has been possible to establish cultures of large granular lymphocytes in IL-2, following their separation on discontinuous Percoll gradients. These cultured lymphoid cells maintain the morphology and function of native NK cells. Cultured lymphoid cells specific for human tumors have also been derived after in vitro sensitization and separation of T-cell blasts from large granular lymphocytes (LGL) on similar density gradients. In a study described in the chapter, the optimal conditions for initiating IL-2 dependent cultures from LGL and small T cells were examined by limiting dilution analysis. The role of interferon in modulating IL-2-driven population expansion and the mechanism of that effect were also studied, using the limiting dilution analysis. The sensitization in MLR does not decrease but rather increases the frequency of NK cell precursors and provides newly activated T cells." @default.
