The method is based on the oxidation of succinate by succinate dehydrogenase, followed by oxidation of the reduced enzyme by Phenazine Methyl Sulfate (PMS) and cytochrome c. PMS acts here as an electron carrier between enzyme and cytochrome c. Although the reduced form of PMS is very autoxidizable, oxygen does not affect the determination. 2, 6-Dichlorophenol indophenol can be used instead of cytochrome c, but the reported values for the molar extinction coefficient show a large variation. Perchloric acid in a final concentration of 5% is added to the sample. The sample is centrifuged, and potassium carbonate solution is added to a portion of the supernatant up to pH 7-8. The mixture is allowed to stand at 0° for 15 minutes and centrifuged at 0°. The supernatant is used in the assay." @default.
