- W979989813 abstract "The most effective, as well as the simplest, method of predicting chemical reactivity or biological activity as a function of molecular structure is correlation analysis (frequently termed linear free energy relationships). Its application to the prediction of prodrug lability has been described. The correlation equations for the appropriate reactions can be used to predict whether the proposed prodrugs will undergo significant cleavage before reaching the site at which they are to act and whether they will form the parent drug at a suitable rate. When the path of the prodrug from site of entry to site of action is fully known, the lability of the prodrug is predictable provided the necessary correlation equations are known. The correlation equations can be used to make qualitative estimates of lability that can be useful both in the choice of a reversible group and in the selection of the most suitable derivatives of a reversible group." @default.
