Purification involves preparation of membrane pellet; solubilization of pellet with NP-40; ammonium sulfate precipitation; DE-52 ion-exchange chromatography; and rechromatography on DE-52. The enzyme is collected by centrifugation and stored in 50% saturated ammonium sulfate at 4°. Assay method is based on the principle that states glucosidase-I is active with the substrates Glc3-oligosaccharide-α-lactalbumin, Glc3-oligosaccharide-lipid and Glc3-oligosaccharide. For ease of assay, the use of [14C-Glc]3 oligosaccharide-α-lactalbumin is recommended. The synthesis of glucosylated α-lactabumin is The enzyme is assayed by measuring the release of [14C]glucose after removal of the protein by TCA-PTA precipitation.
