- W988112996 abstract "Publisher Summary Significant observations concerning cellular activities were made soon after autoradiography was introduced as a cytochemical tool, autoradiography in its infancy did not escape the inherent problems that usually accompany a new technique. In recent years, the field of autoradiography has expanded rapidly because of the availability of tritiated compounds of very high radiochemical purity and the production of fine-grained nuclear emulsions. Investigators now have a more critical means for introducing radioactivity into specific cellular components in view of the many tritiated precursors that can be applied to biological investigations. Autoradiographic patterns are analyzed more accurately, because the short range of tritium radiation permits a resolution of 1μ with conventional methods at the light microscope level. The novice in electron microscope autoradiography will be informed about the important contributions that light microscope observations can make to high-resolution autoradiography, the incorporation of several steps in electron microscope autoradiographic experiments that will make operations more easily and quickly performed, and the use of a simplified method for obtaining suitable electron microscope autoradiographs that will provide adequate resolution for critical evaluation." @default.
