Despite recent exciting discoveries of inherited susceptibilities to cancer, such as the association of certain colon cancers with a deficiency in mismatch repair (23; see Chapter 20), Doll has argued that exogenous agents and lifestyle continue to play a major role in determining human cancer incidences (20). For this reason, the mechanism of action of chemical carcinogens remains an important area of research. Although it has been argued that chemicals can induce carcinogenesis in the rodent bioassay system through either a mitogenic or mutagenic mechanism (1), most of the known potent carcinogens are strong mutagens, and it is these genotoxic carcinogens that are the subject of this chapter." @default.
