- W997215846 abstract "Publisher Summary This chapter discusses thermal measurements during oxidative reactions, and describes the design and operating characteristics of a differential calorimeter suitable for measurement of enthalpy changes during metabolism by fragile subcellular particulates such as mitochondria. A differential calorimeter has been constructed, consisting of two dewar flasks, a sample and a reference, whose temperatures are compared with a thermopile. The 50 ml dewar flasks are wrapped in aluminum foil and placed in hollow aluminum cylinders, which are closed at one end. The block diagram of the electronic circuitry associated with the calorimeter is given. The thermopile voltage is amplified by a Leeds and Northrup DC microvolt amplifier (Model 8739-A). The thermopile output, per copper-constantan thermocouple, is 40 microvolts per degree temperature difference between the two dewars. The calibration of the specific heat of the sample dewar can be measured by two different methods: (1) ohmic heating with a precision resistor, and (2) the neutralization of a known amount of a strong acid by a strong base. The specific heat of the dewar was measured to be slightly larger than the specific heat of the liquid contents of the dewar. This calorimetric method has been applied to a study of the energy balance during oxidative phosphorylation by liver mitochondria.
