- W99807671 abstract "In an earlier in vitro study we observed that high doses and dose rates of I-131 induced DNA single strand breaks (ssb) in human mononuclear blood cells. However, the relatively small doses and dose rates of I-131, which are present in the blood of thyroid cancer patients after treatment with 4-7.5 GBq I-131 (i.v.), caused an average 32% decrease in DNA single strand breaks (40 patients; p < 0.01). A possible explanation for the decrease in spontaneous ssb could be a radiation induced stimulation of DNA repair enzymes. Therefore, in the present study we investigated the DNA repair capacity of human mononuclear blood cells under continuous treatment with I-131 in vitro using a modified alkaline elution method. Mononuclear blood cells were incubated with 22 MBq I-131/ml at 4{degrees}C and 37{degrees}C, respectively, for 100, 200, 300, 400, and 500 min. At 4{degrees}C, where DNA repair enzymes are almost inactive, a linear, time dependent increase in ssb was detected (1140 ssb/cell/100 min). At 37{degrees}C an increase of only 330 ssb/cell was observed after 200 min and of 750 ssb/cell after 300 min compared to control cells. Our results show that during an initial interval (0-300 min of incubation at 37{degrees}C) the number of induced ssb exceeded the number of repaired ssb. During the following interval (300-500 min) more ssb were repaired than induced.
