- W998416130 abstract "This chapter presents the measurement of binding of nonheme ligands to hemoglobins. Most hemoglobins are very sensitive to changes in environmental conditions although in some lower vertebrates and invertebrates, oxygen carrying proteins are found that appear to be unaffected by variations in cellular composition. In general, the oxygen-carrying ability of hemoglobin is modulated by components of the solvent, the so-called “third components.” In other words, the regulation and control of hemoglobin function involve binding of solvent components at sites different from the heme group. Chemical entities that affect the oxygen-binding properties of hemoglobin are known as “oxygen-linked molecules,” and their effect is termed as “heterotropic linkage” as opposed to homotropic linkage, which involves interactions among the heme groups themselves. Thus, the response of the hemoglobin molecule to changes in environmental conditions involves interactions between heme and nonheme binding sites, transmitted by the protein moiety. The quantitative evaluation of the various linkages existing between the different ligands of the protein may provide the basis for a better understanding of the complex molecular mechanism of heterotropic effects.
